Herpes simplex virus type 1 (HSV-1) is a neurotropic human pathogen that establishes a long-term interaction with the infected host. After the primary infection, the virus ascends into sensory nerve axons, leading to latent infection and periodic reactivation. This phenomenon affects most HSV-1 patients and is closely associated with the host’s immune status. Among HSV-1 proteins, ICP27 (Infected cell protein 27) is a multifunctional protein encoded by gene UL54 (Unique Long region 54) that plays an essential role in viral infection, acting on multiple levels to regulate cellular homeostasis and immunity in favor of the virus, ensuring its survival and replication. 

This study demonstrates that ICP27 downregulates gene and protein expression of the Absent-in-melanoma-2 (AIM2) inflammasome components. Consequently, this reduces the production and release of the pro-inflammatory cytokines IL-1β and IL-18, impacting the activation of the innate immune response and potentially weakening the protective response to infection. These findings offer valuable insights for developing effective vaccine strategies to counter herpetic infections.

Dopo l’infezione primaria, il virus risale negli assoni dei nervi sensoriali, portando a un’infezione latente e a una riattivazione periodica.
